What are Polycomb group proteins doing in mouse limb development?

 

講演内容

Polycomb group (PcG) proteins play a pivotal role in silencing developmental genes and help to maintain various stem/precursor cells and regulate their differentiation. PcG factors also regulate dynamic and complex regional specification, particularly in mammalian tissue/organ development. However, this activity is not mechanistically well understood due to the varieties in PcG factors and embryonic lethality in knockout mouse. In this seminar, I will present our recent findings on the function of PcG factors in organogenesis using mouse forelimb bud, a suitable experimental model system for understanding how organogenesis is orchestrated along three axes, that is, anterior-posterior, dorsal-ventral and proximal-distal axes. By combining the conditional knockout mouse system, transcriptome and chromatin immunoprecipitation analyses, we have found that loss of PcG proteins, such as RING1A and RING1B, induced a severe degects in forelimb formation along the proximal-distal (PD) axis and that RING1A/B are essential for PD specification of the forelimb bud via repressing Meis genes in the distal region of forelimb bud. Furthermore, we propose that PcG factors are required to adjust the threshold for Retinoic acid (RA)-related signaling to regulate Meis expression. Taken together, we have discovered a role for PcG factors in repression of Meis expression by creating de novo PcG-repressive domains through antagonizing RA-related developmental signals.
