ウシの受精卵がゲノム統合に失敗し、発生が停止するメカニズムの一端を明らかにしました。受精後、父由来と母由来のゲノムはそれぞれ「前核」を形成し、細胞の中央で融合(ゲノム統合)する必要があります。本研究では、この過程で「中心体」から伸びる微小管が、モータータンパク質ダイニンを前核膜へ適切に配置させる司令塔として機能していることを突き止めました。特に、2つの前核の間に中心体が位置することが、前核同士を引き寄せ、正常な発生を導く鍵であることを解明しました。

不妊治療において卵子の体外成熟技術は重要ですが、体外で育った卵子は体内由来のものに比べて発生能が低いという課題があります。本研究では、NAD(H)の前駆体であるニコチンアミド・モノヌクレオチド(NMN)を成熟培地に添加することで、卵子の質が向上することを明らかにしました。NMNの添加により卵子内のNAD(H)レベルが上昇し、それに伴ってミトコンドリアの機能に関連する遺伝子群の発現が変化しました。その結果、ミトコンドリアの活性が高まり、エネルギー源であるATP産生量が増加する一方で、細胞にダメージを与える活性酸素(ROS)が減少しました。さらに、減数分裂時における染色体の分配異常(染色体ラグ)も抑制されることが確認されました。これらの相乗効果により、体外受精後の胚盤胞への発生能が有意に改善しました。本成果は、生殖医療における新たな技術展開に貢献することが期待されます。
